  • How much DNA template do I need to give to BATJ?

    BATJ recommends the following quantities for each sequencing reaction. Higher DNA concentrations typically result in stronger signal intensity.

    Plasmid Template:

    Single-stranded 150 – 300 ng
    Double-stranded 300 – 800 ng (depending on insert and vector size)

    We recommend doubling the above amounts in case a repeat sequencing reaction is needed.

    What is required for premixed samples?
    1. In 7 µL total volume, add 400 ng of double-stranded DNA or 50 ng of cleaned PCR product.
    2. Include 3.2 pmol (about 20 ng) of primer in the 7 µL volume.
    3. Please leave a note indicating that your samples are premixed.

    • 1. How about Nucleic Acid Isolation?
      • Q: What types of sample can you do nucleic acid isolation?
        A: We have extensive experience with FFPE tissue samples, microbiota, microbial cultures, cells and tissues (human/animal/plant), environmental samples, and custom samples.
      • Q: What method do you use for nucleic acid extraction?
        A: We use a variety of commercial kits or in-house protocols developed for specific sample types.
      • Q: Can you isolate microRNA?
        A: Yes. We isolate genomic DNA, total RNA, mRNA, and small non-coding RNA, including microRNA.
      • Q: How do you check the quality and quantity of nucleic acids?
        A: Using a fluorometer or Bioanalyzer.

    • 1. Do I have to provide primers (oligos)?

      2. Uncommon primers:

      Customers should provide uncommon primers at a concentration of 10 pmol. BATJ can dilute them if needed.
      BATJ also offers synthesis of primers for an additional charge.

    • 1. How do I place an order and schedule a pickup?

      Please download the request form for the desired service, fill it in, save it, and email it to:

      Also, please give us a call in case we do not receive your email due to internet issues.

      Note: All orders (except PCR orders) placed by 4:00 PM local time will be picked up on the same day.

    • 2. By what time do we need to submit PCR orders?

      To receive PCR sequencing results by the next day, please submit PCR orders before 12:00 PM (noon). These orders will be picked up at noon to allow time for PCR, clean-up, and sequencing.

    • 3. Does BATJ accept mail-in orders?

      Yes. Orders from locations far from our base can be mailed via UPS or FedEx for timely delivery to:

    • 1. How soon can I receive the results?

      Generally, results will be available the next day, no longer than 24 hours.

      For orders including services such as primer synthesis, sample purification, or primer walking, the turnaround time may vary depending on the service.

    • 2. How will I receive the results?

      There are two delivery methods:

      • Small orders are sent via email.
      • Large orders may be transferred through FTP.
    • 3. What format will the files be in?

      Sequencing results are provided as plain text files and electropherogram (.ab1) files.

    • 4. Where can I get the necessary programs to read the sequencing data?

      If you don't have commercial software like ABI Sequencing Analysis, the following free tools are available:

    • 5. What is your repeat policy?

      As of 2017, BATJ does not offer free repeats by default. However, if a sequencing failure is suspected to be BATJ’s responsibility, we will reuse the same DNA and primer for a one-time repeat. If the issue persists, the additional attempt will be treated as a new order.

    • 1. How long do you keep my samples?

      BATJ does not retain any customer sequencing data.

      We store DNA samples for one week in case a reaction needs to be repeated.

      If you would like us to retain your DNA or primers for future orders, please include a note in the “Comments” section of your order such as:

      “Please keep my DNA, primer, or both for future orders.”

      In such cases, we will store your DNA and/or primers for up to one month.

    • Gene Codes Corporation offers a demo of their "Sequencher" program for many platforms. The demo lacks some features.
    • The James Hutton Institute offers free software called Tablet, available for Windows (32 & 64 bit), Linux (32 & 64 bit), and Mac OS X to read NGS analyzed data.
